Sampling Methods
1. Diatoms from the intensively sampled pools were collected two times a year (May and November) from 1978 to 2022, along with records of water level and samples for surface water chemistry. In the extensively sampled pools, diatoms were collected in summer or early autumn every four years from 1978 to 2012, along with chemical samples. 2. Samples were taken with a plankton net (mesh size 30-40 µm) pulled slightly through macrophytes if present, and slightly through the upper sediment layer. Until 2010 the samples were preserved with formaldehyde (4%), later on the samples were stored at -12 °C. 3. In addition to these 'recent' samples, net samples collected by J. Heimans in the years 1916 - 1932 (now in Naturalis Biodiversity Centre, Leiden) were examined, supplemented with samples from later years from employees of the former Research Institute for Nature Management (now in Meise Botanic Garden). We have made all efforts to ensure the comparability of our sampling method with the historical sample methods. 4. In the laboratory, HCl (10%) was added to remove the diatoms from the macrophyte fragments and to dissolve iron (when present). After rinsing several times with H2O, the organic material was digested by warming (80 °C) in H2O2 (30%). From 2014 onwards, KMnO4 was added after this step to end the oxidation process. After rinsing several times with H2O, slides were prepared by embedding the diatom valves in Naphrax. The slides were examined under oil immersion using a Zeiss Standard RA or Axioskop 40 microscope and phase contrast objective (x 100, NA = 1.30, total magnification x 1250). 5. During the first years of the project, diatoms were identified using van der Werff & Huls (1957-1974) and Hustedt (1927-1966, 1930) as main reference works and special publications as listed by van Dam (1984). Krammer & Lange-Bertalot (1986-1991) was used as a central reference in the following decades, supplemented by many special publications, as listed in van Dam (1984) and van Dam & Mertens (2019). The taxonomy in the checklist by van Dam et al. (1994) is used as a base-line in the present study, with some exceptions: Eunotia bilunaris var. mucophila Lange-Bertalot et al. is included in E. bilunaris (Ehrenberg) Mills, E. steineckii Petersen is included in E. exigua (De Brébisson) Rabenhorst, Frustulia rhomboides var. crassinervia (De Brébisson) Ross is included in F. saxonica Rabenhorst, Navicula heimansioides Lange-Bertalot is included in N. leptostriata E.G. Jørgensen. The group around N. subtilissima Cleve is combined to the genus Kobayasiella Lange-Bertalot. The nomenclature is adapted to the most recent insights (Guiry & Guiry, 2021). 6. In each slide, 400 valves were counted. The per cent abundance of each taxon was calculated after counting.
